To define a “Hotkey” for any key, double-click or click an item in the list and hit the “Return” key. Next, type the “Hotkey” combination you'd like to assign to this key.
Example, let's say you wanted to set the function key F12 to be the Forward Delete key. Double-click the item in the list named “Forward Delete”. Now hit the 'F12' key to set the “Hotkey” and make sure the “On” check box is checked. Click the “OK” button.
Once you close the KeySwapper window (and if you have KeySwapper installed in the System Folder), hitting the F12 key will make the system believe you actually typed the Forward Delete key!
Here are the default “hotkeys” and the keys they emulate:

Left Arrow = Option-Keypad 4
Right Arrow = Option-Keypad 6
Up Arrow = Option-Keypad 8
Down Arrow = Option-Keypad 2
Caps Lock Toggle = Control-`
You can turn any of these off at any time or change the “hotkeys” at any time by opening the KeySwapper control panel and double-clicking the item you wish to change.
